AFFINIMIP® SPE 18F - Aromatic Nucleophilic Substitution

Purification of 18F radiotracer obtained from ammonium precursor by Aromatic Nucleophilic Substitution.

AFFINIMIP® SPE for purification of radiotracers is based on the affinity for the precursor. In the case of Aromatic Nucleophilic Substitution with 18F, impurities, Kryptofix and residual 18F are also retained on the cartridge, while the purified radiotracer is obtained after elution.

 

 

 

 

 

 

 

AFFINIMIP® SPE 18F - Aromatic Nucleophilic Substitution is a selective solid phase extraction cartridge used to purify rapidly and perfectly a 18F radiotracer synthesized from an aromatic nucleophilic substitution. The cartridge retains the ammonium-based precursor as well as the phenol- and dialkylamino-based impurities.

In this article, the authors describe the automated synthesis of [18F]FPCA peptide, a novel, water-soluble prosthetic group using AFFINIMIP®SPE 18F - Aromatic Nucleophilic Substitution purification. It's radiochemistry has been developed and fully-automated for application in chemoselective radiolabelling of amino(oxy)-derivatised RI-OR2-TAT peptide, (Aoa-k)-RI-OR2-TAT, using a GE TRACERlab FX-FN. RI-OR2-TAT is a brain-penetrant, retro-inverso peptide that binds to amyloid species associated with Alzheimer's Disease.

In this article, the authors describe the preparative synthesis of [18F]FPyPEGCBT for which its HPLC purification was replaced by AFFINIMIP®SPE 18F - Aromatic Nucleophilic Substitution purification with a DMF elution of the product of interest. The authors conclude that the advantage of such purification are "the obviation of a time-consuming HPLC step and a significant simplication of the overall radiopharmaceutical protocol."
